Gaucher disease is passed down from parents to children (is inherited). It is caused by a problem with the GBA gene. It is an autosomal recessive disorder. This means that each parent must pass along an abnormal GBA gene for their child to get Gaucher.

Where is Gaucher disease found?

Gaucher (go-SHAY) disease is the result of a buildup of certain fatty substances in certain organs, particularly your spleen and liver. This causes these organs to enlarge and can affect their function. The fatty substances also can build up in bone tissue, weakening the bone and increasing the risk of fractures.

Is Gaucher disease more common in one ethnicity?

Gaucher disease is considerably more common in the descendants of Jewish people from Eastern Europe (Ashkenazi), although individuals from any ethnic group may be affected. Among the Ashkenazi Jewish population, Gaucher disease is the most common genetic disorder, with an incidence of approximately 1 in 450 persons.

What is the history of Gaucher disease?

Gaucher's disease was first described by Philippe Gaucher in 1882, who recognized that this was a systemic disease. The biochemical defect, genetic basis and molecular epidemiology have subsequently been characterized.

What genetic mutation causes Gaucher disease?

Mutations in the GBA gene cause Gaucher disease. The GBA gene provides instructions for making an enzyme called beta-glucocerebrosidase. This enzyme breaks down a fatty substance called glucocerebroside into a sugar (glucose) and a simpler fat molecule (ceramide).

How is Gaucher inherited?

Gaucher disease is passed down from parents to children (is inherited). It is caused by a problem with the GBA gene. It is an autosomal recessive disorder. This means that each parent must pass along an abnormal GBA gene for their child to get Gaucher.

Who is most likely to get Gaucher disease?

Gaucher disease can affect anyone, occurring in up to 1 in 40,000 live births in the general population. Gaucher disease is more common among Jews of Ashkenazi (Eastern European) descent, occurring in approximately 1 in 450 within this population.

Who named Gaucher disease?

Gaucher's disease is the most common of the lysosomal storage diseases. It is a form of sphingolipidosis (a subgroup of lysosomal storage diseases), as it involves dysfunctional metabolism of sphingolipids. The disease is named after the French physician Philippe Gaucher, who originally described it in 1882.

How Gaucher's cells are formed?

GCase is activated by saposin C. In lysosomal storage diseases, an enzyme deficiency is responsible for the accumulation of its substrate in the cell lysosome (overload disease). Gaucher disease is caused by a deficiency in glucocerebrosidase (GCase) (or β-glucosidase), which leads to an accumulation of GlcCer.

Is Gaucher disease a disability?

In most cases, claims for Social Security Disability benefits based on a diagnosis of Type 2 Gaucher Disease are quickly approved by the SSA. In some cases, however, even claims based on conditions covered under the Compassionate Allowances guidelines are denied.

What is the life expectancy of a person with Gaucher disease?

From the International Collaborative Gaucher Group (ICGG) Gaucher Registry, the mean life expectancy at birth of patients with Gaucher disease Type 1 has been reported as 68.2 years (63.9 years for splenectomised patients and 72.0 years for non-splenectomised patients), compared with 77.1 years in a reference ...

Is Gaucher disease an autoimmune disease?

Conclusions: Forty five percent of the evaluated type I Gaucher patients exhibited autoimmune phenomena. Additionally, 24% presented with lymphoproliferative disorders. DC function analysis showed a significant impairment of both iDCs and mDCs, reflected by their decreased uptake and antigen presenting capacities.

Can Gaucher disease be cured?

While there's no cure for Gaucher disease, a variety of treatments can help control symptoms, prevent irreversible damage and improve quality of life. Some people have such mild symptoms that they don't need treatment.

How is Gaucher disease prevented?

There is no way to prevent Gaucher disease if you have the gene mutations. It's wise to have testing if you are at risk. Early treatment may prevent damage to bones and organs from Gaucher disease type 1.

What chromosome is Gaucher disease found on?

Gaucher disease, the inherited deficiency of the enzyme glucocerebrosidase (EC3.2. 1.45), is the most common inherited lysosomal hydrolase deficiency. The gene for glucocerebrosidase (GBA) is located on chromosome 1q21 (Ginns et al.

Does Gaucher disease cause weight gain?

Diet as part of overall care for Gaucher disease

People with GD may be at higher risk for certain medical issues, and a nutritious diet can keep them on a healthy path. Weight gain: People often gain weight after starting ERT, which can lead to an increase in fatty liver and a risk of developing fatty liver disease.

How common is Tangier disease?

Tangier disease is a rare disorder with only approximately 100 cases diagnosed worldwide.

Is Gaucher disease fatal?

Because of the devastating brain damage, Gaucher disease type 2 is typically fatal within the first 2 years of life. While Gaucher disease type 2 is currently untreatable, researchers continue to look for answers.

What is the GBA gene?

The GBA gene provides instructions for making an enzyme called beta-glucocerebrosidase. This enzyme is active in lysosomes, which are structures inside cells that act as recycling centers.

What is Hunter syndrome?

Hunter syndrome is a very rare, inherited genetic disorder caused by a missing or malfunctioning enzyme. In Hunter syndrome, the body doesn't have enough of the enzyme iduronate 2-sulfatase.

What causes glucocerebrosidase deficiency?

Glucocerebrosidase deficiency: Causes type 1 Gaucher disease, a progressive genetic disease due to an enzyme defect. The enzyme, glucocerebrosidase, is needed to break down the chemical glucocerebroside. The enzyme defect leads to the progressive accumulation of glucocerebroside in the spleen, liver, and lymph nodes.

What are the physical symptoms of Gaucher disease?

• Enlarged spleen.
• Enlarged liver.
• Eye movement disorders.
• Yellow spots in the eyes.
• Not having enough healthy red blood cells (anemia)
• Extreme tiredness (fatigue)
• Bruising.
• Lung problems.

Is Down syndrome autosomal recessive?

Like cystic fibrosis, Down's Syndrome is autosomal recessive. This means that the condition is genetic and passed down by the mother and/or the father but the condition is not shown in the parents. An autosomal recessive disorder requires two copies of the abnormal gene for the disease or trait to develop.

How is Gaucher disease diagnosed?

An enzyme test called a beta-glucosidase leukocyte (BGL) test is the main tool that physicians use to diagnose Gaucher disease. This is because all patients with Gaucher disease will have low enzyme activity levels. Your physician can measure enzyme activity with a standard blood test.

What claim is being made about the cause of Gaucher disease?

Gaucher disease is caused by changes (mutations) in a single gene called GBA. Mutations in the GBA gene cause very low levels of glucocerebrosidase. A person who has Gaucher disease inherits a mutated copy of the GBAgene from each of his/her parents.
