A hemoglobinometer is an instrument used to determine the hemoglobin content of the blood by spectrophotometric measurement. Portable hemoglobinometers provide easy and convenient measurement, which is particularly useful in areas where no clinical laboratories are available.

What is a hemoglobinometer used for?

A common method for measuring the hemoglobin content of blood makes use of an instrument known as a hemoglobinometer, which compares the colour of light passing through a hemolyzed blood sample with a standard colour. The results of the test are expressed as grams of hemoglobin per 100 ml of blood.

In which method of Hb estimation hemoglobinometer is used?

Gasometric method of estimation of hemoglobin is done by using van Slyke apparatus, It is the most accurate method. However it is time consuming and the complex procedure.

What is the principle of hemoglobinometer?

The Digital Hemoglobinometer System is based on the principle of reflectance photometry. Capillary, venous, or arterial whole-blood sample can be used for the hemoglobin estimation with the requirement of only 8 ml of blood sample.

Why is Cyanmethemoglobin is the most preferred method?

The haemoglobin is estimated with the help of cyanmethemoglobin curve. The advantages of this method are i) error due to subjective visual matching is avoided as spectrophotometer is used and hence reading is precise and reliable, ii) measures all forms of haemoglobin except sulphaemoglobin.

What is the Cyanmethemoglobin method?

The method of choice for hemoglobin determination is the cyanmethemoglobin method (This is a type of colorimetric method). The principle of this method is that when blood is mixed with a solution containing potassium ferricyanide and potassium cyanide, the potassium ferricyanide oxidizes iron to form methemoglobin.

What does Cyanmethemoglobin method measure?

Cyanmethemoglobin method measures all types of hemoglobin concentration except. Venous blood collected with EDTA or double oxalate or capillary blood. Specimen requirement for cyanmethemoglobin method.

How do you use a Hemoglobinometer?

To operate the instrument, the user simply allows a drop of blood to fill the disposable cuvette by capillary action and inserts the cuvette into the instrument. The instrument analyzes the 10 microl sample and displays the results in less than 10 seconds.

What is the purpose of using HCL in Sahli's method?

The principle of Sahli's Method or Acid hematin method is quite easy that when the blood is added to N/10 Hydrochloric acid (HCl), the hemoglobin present in RBCs is converted to acid hematin which is a dark brown colored compound.

What are the different types of Hemoglobinometer?

• Hemoglobincyanide (HiCN) Method. ...
• Vanzetti's Azide Methemoglobin. ...
• “Reagent-Less” Methods. ...
• Non-Invasive Methods. ...
• Sahli´s Method. ...
• Hematology Analyzer. ...
• Blood Gas Analyzer (BGA)

Which instrument is used in oxy hemoglobin method & Cyanmethemoglobin method of Hb estimation?

Hemoglobinometers based on the cyanmethemoglobin/oxyhemoglobin method are available. The instrument has a built in filter and a scale and gives direct reading of Hb in g/dl. They are used in field surveys.

What is the most preferred method of hemoglobin estimation?

Direct cyanmethaemoglobin method² is the most widely used and recommended method by the International Committee for Standardization in Haematology for quantitative estimation of haemoglobin.

What is Drabkin solution?

Drabkin′s reagent is used in measuring haemoglobin from blood samples. It comprises potassium ferricyanide, potassium cyanide and potassium dihydrogen phosphate as components. Potassium ferricyanide oxidizes haemoglobin to methemoglobin and then to cyanmethemoglobin.

Who discovered Hemoglobinometer?

British neurologist William Gowers (1845-1915) invented the haemoglobinometer in 1875. It rapidly tested the amount of haemoglobin in blood by comparing its colour with a reference tube.

What if the hematocrit is low?

A lower than normal hematocrit can indicate: An insufficient supply of healthy red blood cells (anemia) A large number of white blood cells due to long-term illness, infection or a white blood cell disorder such as leukemia or lymphoma.

What is the purpose of the hemolysis applicator?

What is the purpose of the hemolysis applicator? It breaks down red blood cells and releases hemoglobin to the solution.

Can nitric acid be used in Hb estimation instead of HCL?

The main principle of this method is the conversion of hemoglobin into hematin on addition of hydrochloric acid. ... The nitric acid or sulfuric acid cannot convert hemoglobin into hematin and no brown color compound is formed by using these reagents.

Why acid hematin is formed?

This pigment is produced by acid acting upon hemoglobin and is known as formalin pigment or acid hematin. ... These parasites disintegrate erythrocytes in an unknown manner, and liberate an acid hematin-like pigment which is phagocytized by the reticuloendothelial system.

Where is Oxyhaemoglobin formed?

Oxyhemoglobin. Oxyhemoglobin is formed during physiological respiration when oxygen binds to the heme component of the protein hemoglobin in red blood cells. This process occurs in the pulmonary capillaries adjacent to the alveoli of the lungs.

How do you use a Haemometer?

The haemometer is an absolute measuring system by graduation with g/dl reading. The graduated measuring tube is filled up to the bottom graduation line with hydrochloric acid. A quantity of 20 μl of blood is blown into the tube and water is added until the colour of the solution matches the colour of the test rods.

What are two conditions that cause polycythemia?

• Hypoxia from long standing (chronic) lung disease and smoking are common causes of polycythemia. ...
• Chronic carbon monoxide (CO) exposure can also be a risk factor for polycythemia.

What is a Hemometer?

Medical Definition of hemometer

: an instrument for measuring some quality of blood (as hemoglobin content)

How does oxygen bind to hemoglobin?

Each subunit surrounds a central heme group that contains iron and binds one oxygen molecule, allowing each hemoglobin molecule to bind four oxygen molecules. Molecules with more oxygen bound to the heme groups are brighter red.

What are measured in the Gasometric and chemical methods for hemoglobin determination?

gasometric method, the oxygen carrying capacity of the blood is measured with the Vonslyke apparatuas and hemoglobin is estimated indirectly. In the specific gravity method, haemoglobin is calculated from the figure for specific gravity determined by copper sulphate technique.

What is PCV in human body?

The packed cell volume (PCV) is a measurement of the proportion of blood that is made up of cells. The value is expressed as a percentage or fraction of cells in blood. For example, a PCV of 40% means that there are 40 millilitres of cells in 100 millilitres of blood.
