The imidazoles are synthetic fungicides that disrupt ergosterol biosynthesis; they are commonly used in medical applications and also in agriculture to keep seeds and harvested crops from molding.

What does imidazole treat?

An imidazole antifungal used to treat vulvovaginal candidiasis. A broad spectrum antifungal used to treat seborrheic dermatitis and fungal skin infections. A topical antifungal used to treat tinea pedis, tinea cruris, tinea corporis, cutaneous candidiasis and tinea versicolor.

What is the mechanism of action of imidazole?

Mode of Action:

Imidazoles alter the cell membrane permeability of susceptible yeasts and fungi by blocking the synthesis of ergosterol (demethylation of lanosterol is inhibited), the primary cell sterol of fungi. The enzyme targeted is a fungal cytochrome P450 (CYP450).

What is the target for the antifungal drugs?

Antifungal drugs target structures or functions that are necessary in fungal cells but not in human cells, so they can fight a fungal infection without damaging your body's cells. Two structures that are commonly targeted are the fungal cell membrane and the fungal cell wall.

What do azoles target?

Azole drugs are broad-spectrum drugs and treat fungal infections of the skin or mouth. An example of an azole drug is Clotrimazole, commonly used to treat athlete's foot, ringworm, vaginal yeast infections, and oral thrush.

What are ‘targeted’ cancer drugs?

Are azoles fungistatic or fungicidal?

Undoubtedly, azoles have divergent effects depending on the fungal species. They exert generally a fungistatic activity against yeasts, e.g., Candida spp., while being fungicidal for certain medically important molds, e.g., Aspergillus spp^3,12.

What is an azole drug?

Azoles are synthetic antifungals with broad-spectrum fungistatic activity against yeasts and fungi, including candidal species. By blocking fungal cytochrome P450-dependent enzymes, azoles disrupt the synthesis of ergosterol, which is the principal sterol in fungal cell membranes.

Why is ergosterol a good target for antifungal drugs?

Ergosterol, a 5,7-diene oxysterol, is the most abundant sterol in fungal cell membranes, where it regulates permeability and fluidity (1). Because of its crucial functions, unique structural properties, and particular biosynthetic steps, ergosterol is the target of the majority of clinically available antifungals (2).

What is a common target of fungal infections?

Primary molecular targets for antifungal agents are enzymes and other molecules involved in cell wall synthesis, plasma membrane synthesis, fungal DNA and protein synthesis, cellular function-related, and virulence factors. The cell wall is a very essential structure of fungi and absents from the mammalian host.

What are imidazole derivatives?

Many imidazole-based derivatives have been marketed as antifungal drugs such as ketoconazole (1), miconazole (2), clotrimazole (3), tioconazole (4), econazole (5), tinidazole (6), enilconazole/imazalil (7), parconazole (8), eberconazole (9), lanoconazole (10), fenticonazole (11), bifonazole (12), sulconazole (13), ...

What is most probable mechanism of action of imidazole antifungal drugs?

A mechanism of action via inhibition of DNA synthesis is an alternative explanation. The imidazole derivatives inhibit the biosynthesis of ergosterol, the main sterol in membranes of fungi. These agents also affect the synthesis of triglycerides and phospholipids.

Which antifungal is safe during pregnancy?

Imidazoles are considered safe as topical therapy for fungal skin infections during pregnancy. Nystatin is minimally absorbed and is effective for vaginal therapy.

Is itraconazole an antibiotic?

Itraconazole is an antifungal medication that is used in adults to treat infections caused by fungus. This includes infections in any part of the body including the lungs, mouth or throat, toenails, or fingernails.

Which antibiotics treat fungal infections?

• clotrimazole (Canesten)
• econazole.
• miconazole.
• terbinafine (Lamisil)
• fluconazole (Diflucan)
• ketoconazole (Daktarin)
• nystatin (Nystan)
• amphotericin.

Which type of fungal infection is most serious?

The most common types of fungi that cause serious or life threatening infections include: Aspergillus, which causes aspergillosis. It most often affects people with lung disease or a weakened immune system. Candida, which causes candidiasis, also called thrush.

What is the mechanism by which most antifungals inhibit fungal infections?

The mechanisms of action of antifungals include inhibition of fungal membrane and cell wall synthesis, alterations of fungal membranes, effects on microtubules, and inhibition of nucleic acid synthesis. Many of the systemic antifungals are toxic to mammalian cells, especially amphotericin B.

Which of the following antifungal agent is having imidazole heterocyclic as a nucleus?

Clotrimazole is an imidazole with broad-spectrum activity against Candida spp.

Why is ergosterol important in fungus?

Ergosterol is a sterol that resides on the cell membranes of fungi and acts to maintain cell membrane integrity, similar to mammalian cholesterol. ... The bound drug molecules form a pore in the ergosterol which allows electrolytes and small molecules to leak out of the cell.

Why is ergosterol important in fungi?

Ergosterol is an essential component of fungal cell membranes that determines the fluidity, permeability and activity of membrane-associated proteins. Ergosterol biosynthesis is a complex and highly energy-consuming pathway that involves the participation of many enzymes.

Why is ergosterol called provitamin D?

Because many fungi and protozoa cannot survive without ergosterol, the enzymes that synthesize it have become important targets for drug discovery. In human nutrition, ergosterol is a provitamin form of vitamin D₂; exposure to ultraviolet (UV) light causes a chemical reaction that produces vitamin D₂.

Which azole is most hepatotoxic?

The azole fluconazole and voriconazole showed a higher hepatotoxic potential; anidulafungin showed in higher concentrations (5x Cmax, 10x Cmax) a severe impairment of hepatocyte vitality, viability, and function.

Is amphotericin B an azole?

The azole antifungal drugs are one example of the fruits of these efforts. Ketoconazole, fluconazole, and itraconazole have ex- tended the ability to treat a variety of fungal infections with oral regimens (9, 11), without the toxicity of amphotericin B.

How do Echinocandins work?

Echinocandins work by inhibiting a glucosyltransferase enzyme (1→3)-β-D-glucan synthase, which is essential for the generation of (1→3)-β-D-glucan, an essential component that maintains the integrity of the fungal cell wall.
