The low-melting mixture urea-ZnCl₂ as reaction medium efficiently catalyzes the formation of imidazoles from a dicarbonyl compound, ammonium acetate, and an aromatic aldehyde to provide a broad range of triaryl-1H-imidazoles or 2-aryl-1H-phenanthro[9,10-d]imidazoles in very good yields.

What is imidazole made of?

imidazole, any of a class of organic compounds of the heterocyclic series characterized by a ring structure composed of three carbon atoms and two nitrogen atoms at nonadjacent positions. The simplest member of the imidazole family is imidazole itself, a compound with molecular formula C₃H₄N₂.

Is imidazole a functional group?

Histidine, an essential amino acid, has as a positively charged imidazole functional group. The imidazole makes it a common participant in enzyme catalyzed reactions. The unprotonated imidazole is nucleophilic and can serve as a general base, while the protonated form can serve as a general acid.

What are imidazole derivatives?

Many imidazole-based derivatives have been marketed as antifungal drugs such as ketoconazole (1), miconazole (2), clotrimazole (3), tioconazole (4), econazole (5), tinidazole (6), enilconazole/imazalil (7), parconazole (8), eberconazole (9), lanoconazole (10), fenticonazole (11), bifonazole (12), sulconazole (13), ...

How many pi bonds are in imidazole?

So, imidazole has two π electrons from the left and right double bonds each. Also, as it turns out, the lone pair on the bottom nitrogen IS within the ring, making it 6 electrons.

IMIDAZOLE (SYNTHESIS, REACTIONS AND MEDICINAL COMPOUNDS)

Where does imidazole come from?

Synthetic imidazoles are present in many fungicides and antifungal, antiprotozoal, and antihypertensive medications. Imidazole is part of the theophylline molecule, found in tea leaves and coffee beans, that stimulates the central nervous system.

Which drugs are imidazole?

The commonly known imidazole compounds are clotrimazole, ketoconazole, and miconazole. Azole derivatives (e.g., fluconazole, itraconazle, voriconazole, ravuconazole, posoconazole) with 1, 2, 4-triazole ring in place of imidazole have better pharmacokinetics profile with better fungicidal activity and lower toxicity.

At which position would imidazole be protonated by acid?

Protonation of gaseous imidazole is predicted to occur exclusively at the N-3 imine nitrogen to yield a stable aromatic cation. Proton affinities in imidazole are calculated as 941, 804, 791, 791 and 724 for the N-3, C-4, C-2, C-5 and N-1 positions, respectively.

What is the example of Diazole?

Diazole refers to either one of a pair of isomeric chemical compounds with molecular formula C₃H₄N₂, having a five-membered ring consisting of three carbon atoms and two nitrogen atoms.

How do I ring my imidazole number?

The carbon atom between the two ring nitrogen atoms is numbered 2 (as in imidazole), and the carbon atom next to the τ nitrogen is numbered 5. The carbon atoms of the aliphatic chain are designated α and β. This numbering should also be used for the decarboxylation product histamine and for substituted histidine.

Is Oxazole basic?

Oxazole is a weak base; its conjugate acid has a pK_a of 0.8, compared to 7 for imidazole.

How do you make thiazole?

Thiazoles were obtained in good yields by the reaction of 1H-1-(1′-alkynyl)-5-methyl-1,2,3-benziodoxathiole 3,3-dioxides with thioamides.

How do you make quinoline?

Quinolines may be obtained by the Skraup synthesis. This involves heating aniline or a substituted aniline with glycerol in the presence of sulphuric acid, ferrous sulphate and nitrobenzene. The reaction is complex. It involves dehydration of glycerol by means of sulphuric acid to give acrolein.

How is furan synthesized?

Synthesis of furans

One of the simplest synthesis methods for furans is the reaction of 1,4-diketones with phosphorus pentoxide (P₂O₅) in the Paal–Knorr synthesis. The thiophene formation reaction of 1,4-diketones with Lawesson's reagent also forms furans as side products.

What is the target of imidazole?

The imidazole derivatives inhibit the biosynthesis of ergosterol, the main sterol in membranes of fungi. These agents also affect the synthesis of triglycerides and phospholipids.

Why is imidazole Basic?

Imidazole is an aromatic ring in the side chain of the amino acid histidine, which is present in nearly all proteins. ... However, imidazole is about 100 times more basic than pyridine. The increased basicity results from resonance stabilization of the charge to both nitrogen atoms.

Is double ring structure with an imidazole ring?

Definition of purines:- It is a heterocyclic aromatic organic compounds with a chemical formula of C5H4N4, it is consists of two rings pyrimidine and imidazole, it includes two of the bases in DNA and RNA, it is water soluble, purines are found in high concentration in meat and meat products, plant based diets are low ...

What drug contains imidazole nucleus?

It is also present in the structure of many natural or synthetic drug molecules, that is, cimetidine, azomycin, and metronidazole. Imidazole-containing drugs have a broaden scope in remedying various dispositions in clinical medicine [1].

How does imidazole elute his tag?

Typically, a low concentration of imidazole is added to both binding and wash buffers to interfere with the weak binding of other proteins and to elute any proteins that weakly bind. His-tagged protein is then eluted with a higher concentration of imidazole.

Do you expect imidazole to be aromatic?

Imidazole - aromatic or not? ... Lastly, imidazole has 6 π - electrons (4 π- electrons from 2 π bonds and 2 π - electrons from the lone pair of electrons of -NH nitrogen atom), i.e., 4n+2 π - electrons where n = 1. Thus, imidazole is an aromatic molecule since it fulfills all of the criteria required to be one.

Is the imidazole ring aromatic?

The imidazole ring is aromatic. Both N atoms are sp2 hybridized.

Would imidazole be a stronger or weaker base than pyrrole?

Imidazole is over a million times more basic than pyrrole because the sp² nitrogen that is part of one double bond is structurally similar to pyridine, and has a comparable basicity.
